The tournament is an official World Ranking event offering Shs78 million in total prize money.
Twenty-one professionals from the Sunshine Tour joined the field of players from eleven nations.
Uganda's Ronald Rugumayo leads the home challenge after winning the Uganda Open and MTN Pro Tee last week.
Other Ugandan professionals include Grace Kasango, Deo Akope and Rodel Gaita.
The opening day features a Pro-Am pairing elite amateurs with professionals for ranking points.
Competition runs from September 17 to 19 under the POA Sunshine Development Tour banner.
Sponsors Exim Bank, Vivo Energy and Uganda Tourism Board back the event.
Uganda Golf Union officials say the strong South African contingent raises the tournament's profile.
